Can never be God's prophet? I'm writing a story for my videogame and need to know this particular detail first.

Yes, provided I get it done. I'm absolutely positive that any Christian will find it quite interesting, as it has an alternate version of history(including the way God's plan works out). I don't want to say any more. ;)

Would having a prophetress be enough to justify allowing woman Pastors?

I like to think of it from this angle. Can God use anyone to fulfill His purpose? If He made a donkey talk, then I don't see why a woman would not be able to speak. If a woman meets the same standard as a man in obtainable pastorial qualities, then I believe that woman should be allowed to be a pastor if she desires.

In contrast, I remember Jesus or Paul saying something about the church leaders being "Husbands of one wife". I think that implies men only...

It shouldn't be overlooked that Paul was also speaking within the culture of his time. I just don't see any reason today why women shouldn't be allowed not to speak. Perhaps in Middle Eastern countries, and other places where men are more respected than women (as like in Paul's time men would be listened to more, and therefore have greater impact). But in Western societies the barriers between men and women have generally been knocked down.
